Analytical Ultracentrifugation Laboratory

 

Two types of experiments are possible in the temperature range of 0 to 40 oC:

• sedimentation velocity – take about few hours (the longer the smaller object is tested),
• sedimentation equilibrium - last up to few days.


Sample volume required: 0.12 - 0.45 ml depending on the type of experiment and cuvette chosen. The required optical parameters:

• Absorption – absorbance at the selected wavelength (190 -
800 nm) in the range 0.1 – 1.5 A with optical path length 0.3 cm or 1.2 cm;
• Fluorescence - emission above 505 nm with excitation at 488 nm;
• Interference at 660 nm.


Experiments with interference and absorption detection may be carried out in parallel on the same sample (the same detector is used), while fluorescence detection requires an independent experiment.
